چکیده

Mice were infected intravenously with 0.5 mg of a virulent strain of M, bovis. The lungs were separated from the moribund mice 3 to 4 weeks after infection. The lungs from 640 mice were homogenized and the bacilli removed therefrom by sequential centrifugation. The supernatant was heated at 120 C for 20 min and subjected to filtration and centrifugation to remove the denatured and aggregated materials. The supernatant was then fractionated with ammonium sulfate at 50 %-saturation, by gel-filtration on a Sephadex G-100 column and by chromatography on a DEAE-cellulose column, The protein fraction finally obtained had a high tuberculin activity and behaved like PPD in disc electrophoresis in polyacrylamide gel.

STUDIES ON THE TISSUE SPECIFICITY OF COLONY STIMULATING FACTOR

In this study granulocyte-macrophage colony stimulating factor (CSF) was prepared from rabbit and guinea pig lung tissues. The biological activity of these CSFs was measured on semisolid agar culture and compared with those of mouse and rat. Weight of animals, incubation time and cross-reactivity of CSFs with different bone marrow sources were also investigated. The results showed that both...

The Identification of proteins present in culture filtrare of Mycobacterium tuberculosis DT and comparison with human tuberculin proteins by electrophoretic methods

Abstract Background and objectives: Tuberculin is the proteins existed in tuberculosis culture medium which precipitated by trichloroacetic acid (TCA) or ammonium sulfate. Tuberculin is used for diagnosis of Tuberculosis. The aim of this study is to compare the human tuberculin produced by Razi Institute and Mycobacterium tuberculosis Culture Filtrate Protein. Material and Methods: Initially By...

Prevalence of Linguatula serrata Nymph in Goat in Tabriz, North-West of Iran

Linguatula serrata is one of well-known members of Pentastomida which infects both humans and animals. The aim of this study was to evaluate the prevalence of L. serrata in mesenteric lymph nodes, livers and lungs of goats slaughtered in Tabriz area, Iran. Mesenteric lymph nodes (MLNs), livers and lungs of 280 goats slaughtered in Tabriz, North-west of Iran were investigated for nymphs of L. se...

Skin test performed with highly purified Mycobacterium tuberculosis recombinant protein triggers tuberculin shock in infected guinea pigs.

Tuberculin shock due to inoculation of Mycobacterium tuberculosis antigens in patients with tuberculosis is a serious syndrome originally described over 100 years ago by Robert Koch. Here, we present experimental evidence that a single M. tuberculosis recombinant protein, CFP-10, triggers this syndrome. Intradermal inoculation of CFP-10 elicits in M. tuberculosis-infected mice high levels of se...

Isolation, Molecular Identification And Genomic Pattern of‌ ‌Mycobacterium Bovis Isolates Collected From Tuberculin-Positive Cattle‌ ‌in Infected Farms of Shiraz

Background: Mycobacterium bovis is the main cause of tuberculosis in cattle. At the global scale and also in Iran, the most frequent currently-in-use method in the detection of infected cattle is tuberculination. Objective: The present study was aimed to improve our genomic knowledge of Mycobacterium bovis population structure in cattle farms of Shiraz. Materials and Methods: Fifty pathologic...
